Libraries can be prepared from 100pg of DNA (compatible with 100pg to 500ng of input DNA). The protocol takes <3.2 hours from enzymatic DNA fragmentation to completion of library preparation. The protocol requires only two tedious bead purification steps and requires <14 minutes of hands-on time. It fragments DNA with low bias, independent of GC content and salt concentration of the sample. Since DNA is fragmented using enzymes, no expensive equipment or special tubes are required.
